MrgprB4 in trigeminal neurons expressing TRPA1 modulates unpleasant sensations

Gentle touch such as stroking of the skin produces a pleasant feeling, which is detected by rare subset sensory neurons that express Mas-related G protein-coupled receptor B4 (MrgprB4) in mice. We examined small populations MrgprB4-positive trigeminal ganglion and dorsal root ganglion, most these were sensitive to transient potential ankyrin 1 (TRPA1) agonist but not TRPV1, TRPM8, or TRPV4 agonists. Deficiency MrgprB4 did affect noxious pain itch behaviors hairless plantar hairy cheek. Although behavior related acetone-induced cold sensing hind paw was changed, unpleasant response acetone application sucrose splash cheek significantly enhanced Mrgprb4-knockout mice well TRPA1-knockout These results suggest sensations cooperation with TRPA1, rather than sensations. Pleasant may modulate on via MrgprB4.
